From what I can gather from http://www.avisynth.org/
this line #ConvertToRGB24 # For TMPGEnc or VFAPI doesn't need to be uncommented unless you have an older version of Tmpgenc right. Because I noticed colors looked sort of weird (I wish I had a better description) on television when I uncommented this line in FitCD

But this is what avisynth.org had to say

Install VFAPI plugin for TMPGEnc
Disable the direct show filters within TMPGEnc and turn off the VirtualDub proxy before frameserving.
Add "ConvertToRGB24" at the end of your AVS-file
Install the [ReadAVS] plugin
Install Huffyuv codec to create a lossless avi in VirtualDub
